PHARMACOLOGICAL ACTIVITIES OF METHANOL EXTRACT OF LEAVES OF HYPERICUM MYSORENSE B. HEYNE

Abstract
Hypericum mysorense belongs to the family Hyperacaceae. It is a large genus of herbs or shrubs with more than 450 species
distributed worldwide. The plants grow widely in temperate regions and are used in folklore medicine as antiviral,
antidepressant, pain reliever in many parts of the world. The aim of the present study was to evaluate the antioxidant,
antidiabetic, anti-inflammatory and antibacterial activities of methanol leaves extract of H. mysorense and to identify the
bioactive compounds by GC-MS analysis. Antioxidant activities such as DPPH˙ radical, superoxide radical (O2 ●-),
hydroxyl radical (OH●), ABTS●+ radical cation, Fe3+ reducing power and phosphomolybdenum reduction assays were carried out. The maximum DPPH˙ radical, superoxide radical (O2●-), hydroxyl radical (OH●) scavenging activities of methanol leaves extract of H. mysorense were 89.30±0.43%, 88.29±0.14%, 56.42±0.35% at 120 µg/mL concentration
and the IC50 values were 30.75 µg/mL, 19.18 µg/mL, 65.45 µg/mL concentration respectively. The maximum ABTS●+
radical cation scavenging activity of methanol leaves extract of H. mysorense was 95.83±0.43% at 30 µg/mL
concentration and the IC50 value was 9.67 µg/mL concentration respectively. The maximum Fe3+ reduction and Mo6+
reduction of methanol leaves extract of H. mysorense were 69.17±0.31% and 74.81±0.38% at 120 µg/mL concentration
and the RC50 values were 48.30 µg/mL and 46.60 µg/mL concentration respectively. The maximum alpha amylase
inhibition of methanol leaves extract of H. mysorense was 79.60±0.22% at 120 µg/mL concentration and the IC50 value
was 23.19 µg/mL concentration respectively. The maximum haemolytic inhibition of methanol leaves extract of H.
mysorense was 41.88±0.45% at 120 µg/mL concentration and the IC50 value was 143.26 µg/mL concentration
respectively. The antibacterial activity of methanol leaves extract of H. mysorense showed maximum zone of inhibition of
22 mm for Proteus vulgaris and for Staphylococcus aureus as 19 mm at 500 µg/mL concentration. GC-MS analysis of
methanol leaves extract of H. mysorense revealed the presence of active compounds such as Quinoxaline,2-isopropyl-3-
phenyl-,4-oxide, Flavone, Pyrimidine,5-ethyl-2-[4-(4-ethylcyclohexyl)phenyl]-exhibiting antimicrobial, antiviral,
antioxidant, antitubercular activities, etc.
