INVITRO AND INSILICO ANTIOXIDANT ACTIVITY OF METHANOLIC LEAF EXTRACT OF CITRUS MEDICA

Abstract
Oxidative stress has become widely viewed as an underlying condition of most prevalent human aliments. The present study aims to evaluate the antioxidant potential of methanolic extract of Citrus medica. Citrus medica L. (Family: Rutaceae), commonly known as ‘citron’ in English and ‘bara nimbu’ in Hindi. Extraction of the active morphological plant part was commenced with 70% methanol by using soxhlet apparatus. The antioxidant activities were conducted via reducing power assay, hydrogen peroxide radical scavenging assay. Preliminary phytochemical screening resulted in alkaloids, flavonoids, saponins, carbohydrates, terpenoids, steroids, and glycosides, were allowed for insilico analysis to know the pharmacokinetics parameters by Qikprop (Schrodinger software) and Molinspiration (ADME and bioactivity score prediction). CMME has shown increase in percent inhibition of free radicals and its IC50 value was found to be 26μg/mL comparable to that standard ascorbic acid (36 μg/mL).Hydroxyl radical’s percent inhibition of CMME was found to be 34μg/mL which was comparable to that standard ascorbic acid (32 μg/mL).The ADME results revealed that the active constituents of the extract have shown zero violation of the Lipinski’s rule indicating the probability of its higher oral bioavailability and TPSA score less than 140 clearly indicating better permeability into the tissues. The result of bioactivity score of GPCR ligand, ion channel modulator, and nuclear receptor ligand, inhibitor activities towards kinase, protease and enzymes indicated that the compounds exhibited active to moderate score towards all the receptors. From the above, methanolic extract of Citrus medica possessed significant anti-oxidant activity.
